The influence of different commercial surfactants on the enzymatic activity of a commercial alpha-amylase from Bacillus licheniformis (Termamyl 300 L) has been studied. As non-ionic surfactants, alkyl polyglycosides (Glucopon® 215, Glucopon® 600 and Glucopon® 650) were studied, as were fatty alcohol ethoxylates (Findet 1214N/23 and Findet 10/15), and nonyl phenol ethoxylate (Findet 9Q/21.5NF). Also, an anionic surfactant, linear alkyl benzene sulfonate (LAS) was assayed. In general, none of the non-ionic surfactants studied, except Findet 10/15, vary substantially the enzymatic activity. Alcohol dehydrogenases (ADH) are a class of enzymes that catalyze the reversible oxidation of alcohols to corresponding aldehydes or ketones, by using either nicotinamide adenine dinucleotide (NAD) or nicotinamide adenine dinucleotide phosphate (NADP), as coenzymes. In this study, a short-chain ADH gene was identified in Bombyx mori by 5'-RACE PCR. This is the first time the coding region of BmADH has been cloned, expressed, purified and then characterized. The cDNA fragment encoding the BmADH protein was amplified from a pool of silkworm cDNAs by PCR, and then cloned into E. coli expression vector pET-30a(+). ABSTRACT Dormancy in lettuce seeds is a process not yet fully understood. High storage temperatures can cause seed dormancy promoting physiological and enzymatic changes. The goal of this study was to investigate the influence of storage period and environments on the quality and dormancy of different lettuce cultivars. We also investigated the biochemical changes. Microbial extracellular enzyme detection on agar plates by means of fluorogenic methylum-belliferyl-substrates. ArchiMer

A rapid and sensitive method to detect the extracellular enzymatic activity of bacteria colonies grown on agar plates is described. Selective agar media supplemented with protein, starch, chitin, Tween-80, etc. are conventionally used to detect biochemical properties of bacteria. It has been experimentally demonstrated with bacteria pure cultures that fluorogenic Methylumbelliferyl (MUF)-substrates are excellent substrate analogues for normally occurring polymers. Based on MUF-substrate hydrolysis the new method provides reliable qualitative estimates of extracellular enzymatic properties of bacteria within minutes using pure cultures as well as agar plates prepared for colony counts.

Dynamique et enzymologie de la digestion chez l'huitre Crassostrea gigas ArchiMer

Feeding experiments carried out on Crassostrea gigas allowed the authors to set up the stages and the timing of the movement of food throughout the digestive tract. Various activities involved in digestion (glycosidases, esterases, and peptidases) were investigated histochemically in the digestive tract in order to localize the sites of absorption and the sites of protein and carbohydrate digestion. An explicative model of the different stages of digestion is proposed.

Seasonal development of bacterial communities in a coastal marine sediment as related to the input of organic material. ArchiMer

The seasonal development of benthic communities was followed in a coastal marine sediment of the Kiel Bight (Baltic Sea; FRG). Total benthic biomass, (ATP), bacterial biomass, overall benthic activity (heat production), and enzymatic decomposition rates of carbohydrate (d-amylase activity) followed a seasonal cycle strongly related to certain events in the sediment. Among these, the input of the phytoplankton blooms in autumn and spring, the accumulation of organic material during winter, and changes in the physical characteristics of the sediment turned out to be most important for the development of benthic biomass and activity. Processes within the benthic community occurred in very short time scales (within days).

The aim of the study was to evaluate the functional ontogeny of the digestive system of Pseudoplatystoma punctifer through the analysis of the activity of the main intestinal (alkaline phosphatase, aminopeptidase N, maltase and leucine-alanine peptidase), pancreatic (trypsin, chymotrypsin, total alkaline proteases, bile-salt activated lipase and amylase) and gastric (pepsin) enzymes. Enzymatic polymorphism study as help for constitution of initial broodstock for a new cultivated finfish species. ArchiMer

Leporinus friderici is a South American promising species for fish culture. In French Guiana the "morphological species", L. friderici, L. granti, L. lebaili and L. affinis steyermarki are found. Their identification is sometimes difficult on usual criterion. To constitute an initial broodstock of L. friderici from local wild populations, the genetic structure of these species was studied. Fifteen enzyme systems, representing twenty loci, were screened.

Proteolytic enzymes of hepatopancreas of six species of peneids (Penaeus kerathurus, P. japonicus, P. monodon, P. stylirostris, P. merguiensis and P. vannamei ) have been studied. Some of their physiological variations have been characterised. Increase in acclimation temperature of crustaceans enhances increase of specific activities of proteases. Variations of salinity does not modify specific activities, nor quantities of trypsin, nor electrophoretic patterns of these different proteases.

The extracellular digestive apparatus of bivalve molluscs consists of a crystalline style containing diverse degradative enzymes which are released during rotation and dissolution of the style. This paper reviews the occurrence of a temperature-regulated bacteriolytic enzyme in the black mussel Choromytilus meridionalis , which is an inhabitant of coastal reefs on the south and south west coast of South Africa. This agent is unable to lyse Micrococcus luteus or Escherichia coli , both of which are susceptible to lysis by a true lysozyme. Different parameters have been followed regularly at various times of anchovy (Engraulis encrasicholus ) ripening. Concentration in NaCl and water content as well as values of water activity and pH are constant and remain quite stable during the entire study period. The relatively acid pH value and the quite low measured water activity level created good conditions for preventing microbial putrefaction at room temperature. The free amino acids and non proteinaceous nitrogen increase indicated the modifications occurring in muscular proteins. ABSTRACT The aim of this study was to evaluate the effect of the essential oil of Lippia alba (EOLA) as a feed additive on ionoregulatory and metabolic parameters and pituitary hormones expression in silver catfish, Rhamdia quelen , submitted to a stressful condition (stocking density of 10.6 kg m-3 and limited space). Fish were fed with different concentrations of EOLA (0.0 - control, 0.25 and 0.50 mL kg food-1) for 20 days. Metabolic parameters were not affected by the diet, with the exception of alanine aminotransferase, which was higher in the liver of fish fed 0.50 mL EOLA kg food-1. Characterization of a leucine aminopeptidase from Toxoplasma gondii OAK

The M17 family leucine aminopeptidase (LAP) hydrolyze amino acids from the N-terminus of peptides. Many LAPs from parasitic protozoa including Plasmmodium, Trypanosoma and Leishmania, have been intensely investigated because of their crucial roles in parasite biology. In this study, a functional recombinant Toxoplasma gondii LAP (rTgLAP) was expressed in Escherichia coli and its enzymatic activity against synthetic substrates for aminopeptidase, as well as the cellular localization was determined. Our results indicated that TgLAP is a functional aminopeptidase in the cytoplasma of T. gondii.

Phosphites for the control of anthracnose in common bean PAB

Abstract: The objective of this work was to evaluate the effect of phosphites in the protection of common bean (Phaseolus vulgaris) against anthracnose. Different phosphite formulations were evaluated by quantifying peroxidase and polyphenol oxidase activity, total phenols, and lignin content. The treatments consisted of sprayings, in the V4, R5, and R7 stages, with: the K, Zn, Mn, K+Mn, K+salicyclic acid, and Cu phosphites; salicylic acid; acibenzolar-S-methyl; the fungicide azoxystrobin; besides a control, without sprayings. A study was carried out to evaluate the effect of broiler breeder age on the development of the digestive tract of embryos and chicks. Fertilized eggs Cobb from 30 and 60 week-old broiler breeder was utilized in this experiment. The results showed that eggs from older (60 weeks of age) broiler breeders were heavier (p = 0.001) than those from younger (30 weeks of age) broiler breeder. The activity of three commercial microbial phytase (Aspergillus oryzae, A. niger, and Saccharomyces cerevisae) products used in broiler nutrition was determined at different pH (2.0 to 9.0) and temperature (20 to 90°C) values. Enzymatic activity was determined according to the reaction of the phytase with its substrate (sodium phytate), in four replicates, and was expressed in units of phytase activity (FTU). A. oryzae phytase exhibited optimal activity at pH 4.0 and 40°C, but its absolute activity was the lowest of the three phytases evaluated. Abstract The changes in concentration of vitamin C, enzymatic and antioxidant activity during the ripening of two soursop selections (G1 and G2) at room temperature (22 ºC) and refrigeration (15 ºC) with an HR 85% were evaluated. The content of soluble protein, the activity of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), the concentration of vitamin C, as well as the antioxidant activity was evaluated by DPPH, ABTS and FRAP methods. The initial soluble protein concentration of the G1 and G2 selections diminished at 22 and 15 °C during ripening. Fruits stored at 22 °C showed the highest CAT activity. Abstract The objective of this work was to validate the enzymatic activity in Annona x atemoyaseeds submitted to treatments with plant growth regulators belonging to the group of gibberellins and cytokinins for overcoming dormancy during the germination process. Initially, the water acquisition curve was determined, where two points of phase II (72 and 144 hours) were determined to evaluate the activity of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) enzymes in atemoya seeds after the application of two plant growth regulators.
